B12B is a promising agent in cellulase enzyme production\r\nthat was affected by several factors, including substrate pre-treatment, \r\nenvironmental conditions and nutrition. Pineapple peel is expected to be an \r\nalternative substrate in the production of cellulase enzymes. This study aims to \r\ndetermine the effect of pre-treatment variations on the pineapple peel substrate \r\n(Ananas comosus L. Merr) and agitation factors, and the optimum concentration of \r\npineapple peel substrate in the production of cellulase by fungi isolate Penicillium\r\nsp. B12B. The stages of this research include physical and chemical substrate pre�treatment, pre-treatment substrate test and agitation factors that were analyzed by\r\nTwo Way ANOVA, and optimization of the substrate concentration of pineapple \r\npeel in cellulase production that analyzed by Paired Simple T-Test. The results \r\nshowed that physical and chemical pineapple peel pre-treatment and agitation \r\nfactors have affected the production of cellulase by fungi Penicillium sp. B12B in\r\ndifferent level, with the highest range of enzyme activity was 0,009 U/mL at 48 to \r\n72 hours. Enzyme activity in the optimization of substrate concentration has a sig.\r\nvalue (2-tailed) of 0.006 < 0.05. Thus it was known that variations in susbtrat \r\nconcentration has affected cellulase enzyme activity by fungi Penicillium sp.
