Most lipases are applied as bio-detergent additives. In this research, aims to isolation of lipase from bacteria isolated from slaughterhouse wastewater and to test its potential as a bio-detergent additives. It also aims to determine how to purify lipase using column chromatography with Ca-bentonite as the matrix. Rhodamine Olive oil Agar (ROA) medium was used for the selection of lipase-producing bacteria. Determination of lipase activity was carried out using the copper-soap calorimetry method. Crude lipase extract was purified using column chromatography with Ca-Bentonite as the matrix and NaCl in phosphate buffer as the eluent. The potential test of lipase as a bio-detergent additive was carried out using stability test exposing by comercial detergent and washing test. The results showed that four isolates were able to produce lipase with each activity of 84.63 U/ml; 114.39 U/mL; 73.88 U/mL; and 62.01 U/mL. Lipase can be purified using column chromatography with Ca-bentonite as the matrix. Crude extract lipase and lipase purified from four isolates was potential as a biodetergent additive except 0.5 M eluent in isolate A2.4.
