Esti Wahyu Widowati, M.Si., M.Biotech %T STUDI POTENSI LIPASE DARI BAKTERI SEBAGAI SALAH SATU KOMPONEN PENYUSUN BIODETERGEN %X Lipase enzymes belong to a class of hydrolase enzymes that are soluble in water and could catalyze the hydrolysis of oils and fats. Lipase could be found in oil-contaminated soil and could be used as a biodetergent. This study aims to isolate bacteria from the landfill soil, then apply the lipase producing bacteria as one of the components of biodetergent. The medium used during the bacterial isolation process was Rhodamine Olive oil Agar (ROA) and the method used when determine the enzyme activity was copper soap colorimetry. Lipase was purified using ion exchange chromatography method with bentonite as the matrix and using NaCl in phosphate buffer as eluent in various concentration. Crude extract lipases and purified lipases are continuously tested against detergent and washing test. Based on the research results, four isolats were obtained with enzyme activity 37.96; 31,732; 34,616; 43.008 U/ml. Lipases that could be applied as one of content of biodetergent are crude extract lipase of S22 isolat and purified lipase from isolat S212 with eluent NaCl 0,1 M and 1 M, isolat S211 with eluent NaCl 1 M, isolat S22 with eluent NaCl 0,5 M, and isolat S32 with eluent NaCl 0,1 M. %K lipase, bentonite, copper soap colorimetry, biodetergent %D 2021 %I UIN SUNAN KALIJAGA YOGYAKARTA %L digilib46952
