This study aims to isolate protease producing bacteria from tempe liquid waste to determine the potential of the resulting protease as a component of biodetergent additives. Isolation of proteolytic bacteria from tempe liquid waste was carried out using skim milk agar (SMA) medium with casein as a substrate. The protease crude extract wast tested for activity using variations in temperature (30, 35, 40, 45, dan 50℃) and pH (6, 7, 8, 9, dan 10). The protease potential of the isolated bacteria as a component of biodetergent additives was tested for stability against expoxure to commercial detergents and a washing test was carried out by adding protein stains to the fabric. Based on the research results from protease isolation, four bacterial isolates were obtained with isolate codes P2a, P6a, P3a (1), dan P4d. Isolate P2a, P6a, and P3a (1) had protease activity of 152,75 U/ml, 92,98 U/ml, 74,75 U/ml, and 49,59 U/ml. Isolate P2a, P6a, and P3a (1) had the highest activity at 50℃ while isolate P4d had the highest activity at 45℃. The four bacterial isolates were able to survive at alkaline pH. The potential of protease as a biodetergent additive with the stability test of enzyme activity obtained a relative percent value above 70% and was able to remove protein stains on fabrics. %Z Pembimbing: Dr.rer.medic.
