Agricultural waste such as dragon fruit stem waste contains cellulose which has the potential to be used as an alternative raw material for bioethanol production. The purpose of this study was to determine the potential of dragon fruit stem waste as a substrate for bioethanol production which was enzymatically hydrolyzed using the cellulase enzyme from Trichoderma viride 6013 and fermented by Saccharomyces cerevisiae 3056. Fermentation was carried out for 96 hours and every 24 hours the levels of bioethanol, pH and dynamics of yeast cell growth were analyzed. The bioethanol content increased significantly with fermentation time with the highest crude bioethanol content obtained in 96 hours of 20% fermentation. The pH decreased from 0 hours at 5 to 4 (72 and 96 hours). Yeast growth was seen to fluctuate with the log phase occurring at 72 hours and starting to enter the death phase at 96 hours.
