This type of research is research and development (Reseach and Development) using the 4-D development model (Define, Design, Develop, and Disseminate). Stages of gathering information through observation, interviews, and documentation regarding invitro conservation of banana cultivars. The research instrument used was a questionnaire sheet. Questionnaire sheets are used for product quality assessment from experts (material experts and media experts), peer reviewers, biology teachers, and student responses. To determine product quality, the assessment was carried out by 1 material expert, 1 media expert, 5 peer reviewers, 1 class X biology teacher at Banguntapan 1 Public High School, and 15 class X students at Banguntapan 1 Public High School. The results of this study were information on in vitro conservation of banana cultivars at the Department of Agriculture and Food of the City of Yogyakarta. The development of a banana cultivar in vitro conservation booklet at the Agriculture and Food Service of the City of Yogyakarta is in the "very good" category with an ideal percentage of 94% from material experts, 98.7% from media experts, 88.3% from peer reviewers, and 80% from biology teachers . The test results of students' responses to the product were 88% in the "strongly agree" category.
