Horticulture industry has high demand availability of orchid species in proportion to their economic value. One of Indonesia's endemic species is Dendrobium lineale, which according to IUCN (International Union for Conservation of Nature and Natural Resources) data is redlisted, meaning it is threatened with extinction. Ex situ conservation efforts that can be done to prevent plant extinction with in vitro culture of seed germination. Media that are often used in orchid in vitro culture and germination are MS (Murashige and Skoog) and VW (Vacin and Went). Additional organic matter in the media is needed to support explant growth. This study aims to determine the optimal composition of MS and VW media with tomato extract additional for the germination of orchid seeds Dendrobium lineale in vitro culture. The study used 4-month-old orchid fruits sown on MS and VW media with additional tomato extract for 18 weeks. The results showed that the treatment of MS media composition with 100 grams of tomato extract was the best media composition for seed germination.
