Seeing this phenomenon, development is needed in the field of PCR-based molecular detection, such as multiplex PCR to detect more than 1 source. This study aims to determine the specificity and sensitivity of the multiplex PCR method using ND5 primers targeting pig and mt-Co1 targeting rat in mixed meat and meatball. The research method has steps, namely DNA isolation, DNA quality & quantity analysis, PCR amplification and electrophoresis. The results of the analysis showed that ND5 and mt-Co1 primers carried out in single and multiple PCR have good specificity and sensitivity capable of targeting pig and rat DNA to the lowest concentration with amplicons of 161 bp and 170 bp in length.
