This study aimed to evaluate the effectiveness of patikan kebo (Euphorbia hirta L.) extract in inhibiting the growth of mastitis-causing bacteria under polar and non-polar conditions, as well as to analyze the effects of solvent type and concentration. The extract was obtained through maceration using polar and non-polar solvents, while Staphylococcus aureus and Escherichia coli isolates were obtained from mastitic milk and characterized using selective media and Gram staining. Antibacterial activity was assessed using the disc diffusion method, determination of Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC), and cell leakage analysis using UV?Vis spectrophotometry. The results showed that patikan kebo extract effectively inhibited both test bacteria, as indicated by the formation of inhibition zones, MIC at a concentration of 40\%, MBC at 50\%, and increased absorbance at wavelengths of 260 nm and 280 nm as indicators of cell membrane damage. These findings indicate that patikan kebo extract has potential to be developed as a natural antibacterial agent for the control of mastitis in cattle.} }
