Abstract

This research aimed to identify hair species in protein contained in bread brush by protein isolation, detection of pig species by porcine detection kit test, and analyzing the proteins in samples with Ninhydrin test and Kjeldahl method. This research used three samples of bread brushes, namely LEX, WNX and TMX trademarks, and used pig hair as positive control and human hair as negative control. The brush was cut into small part and then weighed 0.2 gram, then dissolved into 25 mL NaC12H25SO4 2% solution for 30 minutes. The sample then was added 25 mL Cl2H3K2Na3O8P2 in pH 7.8 and then be incubation at temperature 65oC for 18 hours long. Sample then homogenized using magnetic stirrer for an haour at room temperature. The amount of 42 mL of sampel added by 42 mL H2SO4 10%. The samples were heated in 40oC water bath for an hour and was shaked regularly every 5 minutes. Sample was added 50 mL of NH4HCO3 50%. Samples were detected using Porcine Detection Kit, all of which showed positive results. Ninhydrin test results showed the three samples of bread brushes, pig hair and human hair qualitatively contain protein. Result of Kjeldahl test got average of %N in sample LEX trademark equal to 15.89% b/v, WNX trademark 9.26% b/v, TMX trademark 7.51% b/v, pig hair 7.61% b/v and human hair equal to 7.02% b/v.
