Abstract

Penicillium sp. B12B is a promising agent in cellulase enzyme production that was affected by several factors, including substrate pre-treatment, environmental conditions and nutrition. Pineapple peel is expected to be an alternative substrate in the production of cellulase enzymes. This study aims to determine the effect of pre-treatment variations on the pineapple peel substrate (Ananas comosus L. Merr) and agitation factors, and the optimum concentration of pineapple peel substrate in the production of cellulase by fungi isolate Penicillium sp. B12B. The stages of this research include physical and chemical substrate pre�treatment, pre-treatment substrate test and agitation factors that were analyzed by Two Way ANOVA, and optimization of the substrate concentration of pineapple peel in cellulase production that analyzed by Paired Simple T-Test. The results showed that physical and chemical pineapple peel pre-treatment and agitation factors have affected the production of cellulase by fungi Penicillium sp. B12B in different level, with the highest range of enzyme activity was 0,009 U/mL at 48 to 72 hours. Enzyme activity in the optimization of substrate concentration has a sig. value (2-tailed) of 0.006 < 0.05. Thus it was known that variations in susbtrat concentration has affected cellulase enzyme activity by fungi Penicillium sp. B12B. The optimum substrate concentration in cellulase enzymes production by fungi Penicillium sp. B12B was 2.5% with 0.161 U/mL.
