Abstract

Liquid waste generated from skin cracker production has a fairly high lipid content. The liquid waste from the skin cracker production can increase the toxicity of heavy metals and the high lipid content will interfere with the degradation process on the soil surface. The quality of the soil surface decreases and has a negative impact on the health and environment of the surrounding area. This study aims to obtain lipase from lipolytic bacteria in the liquid waste of skin cracker production and its potential as a biodetergent. Lipolytic bacteria were isolated from the liquid waste of skin cracker production. Lipolytic bacteria are able to grow at hot or thermophilic temperatures because the waste used is in the form of liquid waste at the stage of boiling skin crackers. The medium used in the isolation process is rhodamine olive oil agar (ROA) with olive oil as a substrate. method copper soap colorimetry The isolated lipase was tested for its potential as a biodetergent using the oil displacement area (ODA) and emulsification index methods, and determined the type of surfactant produced by FTIR spectra analysis. Bacteria from liquid waste produced by skin crackers have the potential to produce lipase. Based on the results of the study, there were 4 isolates with isolate codes PK1, PK2, PK3 and PK4 with lipase crude extract activity in the four isolates, namely 109.0; 192.89; 127.69; 91.99 U/ml. The potential of lipase as a biodetergent by determining the values of IE24 (%) and ODA (cm2) obtained the highest activity in isolates PK2 of 41, 26% and PK4 of 19,8 cm2. The type of surfactant obtained from bacterial lipase is rhamnolipid with –OH group at wave number 3400-2400cm-1 and C=O ester group at wave number 1750-1300cm-1.
