Abstract

Isolation of lipase-producing bacteria from the liquid waste of Virgin Coconut Oil (VCO) production was carried out to obtain crude extract of lipase enzyme. Besides being used as a lipid breaker, lipase can be applied as additive components of biodetergents. This study aims to isolate lipase-producing bacteria derived from the liquid waste of Virgin Coconut Oil (VCO) then lipase produced by bacteria is tested for its activity and potential as additive components of biodetergents by testing the stability of lipase activity and washing test. The method stage begins with lipase isolation using Rhodamine B Olive Oil Agar (ROA) media with olive oil as substrats. The method used to determine enzyme activity is copper soap colorimetry using cupric acetate pyridine reagent (CAPR) as dye and olive oil as substrate. Characterization of lipase was performed in pHs (6, 7, 8, 9, and 10) and temperature (30, 35, 40, 45, and 50oC). The crude lipase extract was tested for its stability against exposure to commercial detergents Attack Jaz 1 and a washing test. The results of the study showed that four isolates with isolate codes VC1, VC2, VC3, and VC4 had lipase enzyme activity of 84.63 U/ml; 114.37 U/ml; 106.82 U/ml; and 83.45 U/ml. Four isolates was optimum at pH 10. Iisolates VC1, VC2, and VC3 was optimum at temperature 50oC, while isolate VC4 was optimum at temperature 35oC. The potential of lipase as a biodetergent by testing the stability of enzyme activity and washing test showed that the values of the four isolates were above 70% under exposure to commercial detergents of 7 mg/ml and had a lipid loss percentage exceeding the control percent. Based on the results obtained, lipase from the VCO liquid waste has the potential to be applied as additive components of biodetergents.
