Abstract

Silymarin is composed of a mixture of several flavonolignan, with the most important being silybin, silydianin and silychristin. These compounds has been used as a therapeutic agent in many types of liver diseases, such as hepatitis and cirrosis. With the activity of silymarin, this research is done to isolate silymarin from the seed of Silybum marianum L. Gaertn by effective and simple metode to obtain high levels of silymarin. The study begins with how to deffated Silybum marianum powder with nhexane. Pulp is divided in two and each part was macerated with methanol and ethyl acetate. Partition of the extract are taken twice by adding n-hexane and diethyl ether successively into both of the extracts. Purification was done by recrystallization with two solvent systems, consisted of methanol-water (1:3). Determination of silymarin performed by densitometry using HPTLC (High Performance Thin-Layer Chromatography) plates as stationary phase. The mobile phase used chloroform: acetone: formic acid (9:2:1) and Rf value for silymarin was 0.34, 0.43, 0.57. The results show that silymarin content of methanol extract (53.81%) is higher than the ethyl acetate extract (38.04%). Maximum absorbance of silymarin occured at a wavelength of 288 nm. Analysis of functional groups silymarin with infrared spectrophotometer showed that silymarin in the different prepared samples have functional groups such as: C = O (Carbonyl),-OH (Hydroxyl), C = C (Aromatic), =C–H, and C-O-C (Ether).
