Abstract

Sandalwood (Santalum album L.) has many benefits in various fields, but its existence is endangered. Somatic embryogenesis is an alternative technique for in vitro propagation of sandalwood plants because it is known to be able to produce many embryos in a short time. The purpose of this study was to determine the effect of explant source and optimal media composition to induce somatic embryos and sandalwood embryogenic callus. The study was conducted using RAL with 4 factor groups, with 3 repetitions. The parameters observed were the time of appearance of callus and somatic embryos, the percentage of embryogenic callus and somatic embryos formed, the number of somatic embryos, the fresh weight of embryogenic callus and the morphology and anatomy of somatic embryos and embryogenic callus. The results showed that the fastest time for somatic embryos to appear, the highest percentage of somatic embryos formed and the highest number of somatic embryos occurred in explant sources and different treatment media. Internodal explants and MS+TDZ 3 mg/l media gave quite good results for these three parameters. The somatic embryonic phase found is globular, liver, torpedo and cotyledon phases. Embryogenic callus was produced when leaf explant were treated with MS+NAA 0.01 mg/l+2.4-D 1 mg/l, after 30.5 hst with a weight of 0.0853 grams. Morphologically and anatomically, the resulting callus is embryogenic. Based on the results, it can be concluded that the explant source and zpt concentration added to the media have an effect on the growth of somatic embryos and sandalwood embryogenic callus.
