Abstract

Cemara udang (Casuarina equisetifolia) is one of the species that have the potential as an alternative pulp raw material that is fast-growing and able to adapt to various land conditions. The tree improvement program of cemara udang in Indonesia is currently not widely carried out, therefore the proper cemara udang tissue culture propagation technique needs to be known to encourage the provision of superior seedling clones of cemara udang as an alternative pulp raw material development. This study aims to determine the effect of NAA and BAP hormones and determine the best growth response of cemara udang explants grown on MS and ½ MS media. This study used cemara udang seedlings aged 8 months with a Factorial Complete Randomized Design with 2 treatment factors. The first factor is MS and ½ MS basal media. The second factor is the hormone NAA (0 mg/l, 0.5 mg/l, and 1 mg/l) and BAP (0 mg/l, 1 mg/l, and 2 mg/l). The results showed that the quality of explants was better on ½ MS media than on MS media. Contamination in this study was high and became the main cause of explant death. Browning on the media is 13,3%. In this study, the application of the hormone NAA individually actually eliminated the response of the formation of new shoots and directed more to the growth of calli. The best concentration for shoot induction is in the B1, A2, A1, and B2 treatments, while for callus induction in the A2, A7, and A8 treatments. The sterilization method in this study needs to be reviewed to obtain a high axenic culture.
