Abstract

Research on the analysis of the total phenolic and flavonoid content of teak leaf ethanol extract (Tectona grandis Linn.f) and its correlation with antioxidant activity has been carried out. This study aims to compare the total phenolic and flavonoid content values of teak leaf ethanol extract with varying ethanol solvent concentrations of 50%, 70%, and 96% in maceration extraction and the correlation with the antioxidant activity values expressed by IC values.50. The ethanol extract of teak leaves was taken using the maceration method with varying ethanol solvent concentrations of 50%, 70% and 96%. Determination of phenolic content was carried out using the Folin-Ciocalteu methode by comparing gallic acid and total flavonoid levels using the AlCl3 colorimetric method with quercetin as a comparison. Meanwhile, antioxidant activity testing was carried out using the DPPH method with ascorbic acid as a comparison. The results of this study showed that the highest total phenolic and flavonoid levels were obtained from ethanol extract of teak leaves with an ethanol solvent concentration of 96%. The total phenolic content value of teak leaf ethanol extract with 50%, 70% and 96% ethanol solvent was respectively 684.5; 754.5; and 959.5 mgGAE/g. The total flavonoid content value of young dun teak ethanol extract with 50%, 70% and 96% ethanol solvent was respectively 1032.778; 1105,000; and 1728.778 mgQE/g. The antioxidant activity value of teak leaf ethanol extract with varying solvent concentrations of 50%, 70%, and 96% is expressed as an IC value50respectively 63.53 ± 2.66; 56.09 ± 2.22; and 103.39 ± 16.41 μg/mL. Antioxidant activity value (IC50) of teak leaf ethanol extract with variations in ethanol solvent concentration of 50%, 70% and 96% has a very strong relationship with total phenolic and total flavonoid levels as evidenced by the correlation coefficient (r) 0,9472 for total phenolic content and 0,9862 for total flavonoid content.
