Abstract

One of the charming orchid species is Dendrobium macrophyllum, which is highly sought after due to its pale green color with a labellum that has green stripes and purple lines, and hairs on the back of the petals. Included in Appendix II due to being nearly extinct and slow growth in the wild, thus requiring in vitro orchid propagation. This study aims to determine the effect of Ambon banana and sweet corn extracts on the growth of Dendrobium macrophyllum A. Rich. embryo shoots. The research design uses a factorial completely randomized design consisting of 10 treatments, including 1 control treatment and 9 treatments combining Ambon banana extract: 50; 75; 100 g/L and sweet corn: 50; 75; 100 g/L. The research employs tissue culture, and observations are made every 4th, 8th, and 12th week. The media used were VW (Vacin and Went) media + a combination of Ambon banana and sweet corn extracts. The measured data includes shoot height, number, length, and diameter of leaves, number, length, and diameter of roots, wet weight and dry weight, and shoot morphology. Qualitative data were analyzed descriptively, and quantitative data were statistically analyzed using SPSS. The analysis results show that the best treatment for the parameters of shoot height, leaf length, leaf diameter, and root length is treatment ambon banana 50 g/L and sweet corn 50 g/L. The best treatment for leaf numberand root diameter is treatment ambon banana 50 g/L and sweet corn 75 g/L. The best treatment for root number, wet weight, and dry weight is treatment ambon banana 75 g/L and sweet corn 75 g/L. It can be concluded that the combination of Ambon banana and sweet corn has the potential as a growth medium for Dendrobium macrophyllum A. Rich. embryo shoots.
