Abstract

The association between orchids and mycorrhizal fungi plays a crucial role in plant development, yet the fungal species linked to the roots of Dendrobium macrophyllum remain insufficiently documented. This study aimed to identify fungal isolates associated with wild and cultivated individuals and to analyze their molecular characteristics using the ITS marker region. Fungal samples were isolated through culture, subjected to DNA extraction using the CTAB method, and amplified with ITS1–ITS4 primers prior to sequencing and comparison with reference databases. The results showed that the isolates obtained were not true orchid mycorrhizal fungi but instead belonged to non-mycorrhizal fungal groups closely related to Lasiodiplodia pseudotheobromae. The isolate from wild orchids showed 92.75% similarity, whereas the cultivated isolate displayed 90.64% similarity, indicating genetic variation influenced by differing environmental conditions. Phylogenetic analysis further supported the close relationship between both isolates and the Botryosphaeriaceae family, which is generally characterized by endophytic or saprophytic lifestyles. These findings indicate that the fungi associated with the roots of D. macrophyllum in both sources do not act as mycorrhizal partners but are opportunistic fungi that grow rapidly in culture media. This study provides an important foundation for further research aimed at identifying true orchid mycorrhizal fungi and optimizing mycorrhizal inoculation strategies to support orchid conservation and cultivation.
