Abstract
The genetic diversity of wild bananas (Musa acuminata Colla) adapted to semi-arid regions has potential for breeding drought tolerance. However, studies on these genetic resources are still lacking. This study aims to reveal the level of genetic diversity of several wild banana accessions from Alor Island, East Nusa Tenggara, as a representation of semi-arid regions, and to compare them with accessions from the humid region of Cibinong, West Java, using the molecular marker Sequence-Related Amplified Polymorphism (SRAP). A total of 183 samples from 8 wild banana accessions were extracted for DNA using the CTAB method and PCR with 8 combinations of SRAP primers. The percentage of polymorphic bands from each primer combination was 100% with polymorphic information content ranging from 0.37 to 0.50. The highest Nei’s unbiased genetic diversity was found in the Alor 8.1.2 accession (0.193) and the lowest value was found in the 10H accession (0.091). The total genetic diversity (Ht) was 0.2043, within accessions (Hs) was 0.1369, among accessions (Dst) was 0,0674, with genetic differentiation (Gst) and gene flow (Nm) at 0.3300 and 1.0152, respectively. Analysis of Molecular Variance (AMOVA) showed that 72% of the diversity originated from individuals within accessions and the remaining 28% from between populations. Phylogenetic tree analysis showed that the 8 accessions were divided into 3 clusters, with the first cluster containing the Alor accessions and the second and third clusters containing the Cibinong accessions. Phylogenetic tree cluster analysis and PCoA analysis showed a tendency for accessions to cluster based on their geographical origin.
