Abstract

Liquid tofu waste can serve as a growth substrate for microorganisms in the production of single cell protein (SCP) due to its high carbohydrate and protein content. This study aimed to evaluate SCP production from Saccharomyces cerevisiae FNCC 3049 and a bacterial isolate (R3d) under different fermentation durations (24, 48, and 72 hours), as well as its application as a nutritional supplement in tilapia (Oreochromis niloticus) feed. SCP was produced using the submerged fermentation (SmF) method with tofu wastewater enriched with granulated sugar, (NH₄)₂SO₄, and KH₂PO₄. Results showed that S. cerevisiae achieved optimal growth at 48 hours with 1.92 × 10⁹ cells/mL and 0.8255 g/mL biomass, while the R3d isolate showed optimal growth at 24 hours with 2.813 × 10¹⁰ cells/mL and 0.2206 g/mL biomass. The resulting SCP was incorporated into fish feed and tested in vivo. However, SCP supplementation from both microorganisms had no significant effect on absolute weight gain, survival rate (SR), or feed conversion ratio (FCR) in tilapia. Although SCP was successfully produced and functionally characterized, its application as a feed additive requires further optimization.
