Abstract
Anthracnose disease, caused by Colletotrichum spp., is a major constraint in chili pepper production. The excessive use of synthetic fungicides has spurred the search for environmentally friendly control alternatives. This study aims to explore bacteria from frog skin as a biocontrol agent against Colletotrichum. Four bacterial isolates (RcV2, RcV3, RcV4, and RcV5) were tested using in vitro and in vivo methods. Screening results using the dual culture method showed that isolates RcV2 and RcV4 had the highest inhibition percentages, namely 41.92% and 42.02%. Confirmatory tests using mycelial inhibition assays, conidial germination tests, and slide culture techniques showed that isolate RcV4 at a concentration of 1 × 10⁸ CFU/mL could inhibit fungal mycelial growth with a radius of 23.20 mm and was able to suppress conidial germination by up to 27.27%, as well as induce abnormalities such as dwarfism, fragmented hyphae, and lysis. In vivo test results showed a reduction in disease incidence of 2.23% in fruit and 23.34% in plants. Testing of plant growth parameters, including shoot height, root length, fresh weight (FW), and dry weight (DW), showed significant consistency compared to a commercial fungicide containing 82% mancozeb as the active ingredient. These results indicate that frog skin bacteria have potential as a biocontrol agent due to their antagonistic ability to inhibit the pathogenic fungus Colletotrichum spp.
