Abstract

Chitinase is an enzyme capable of hydrolyzing chitin into simpler compounds and has the potential to be utilized as a biocontrol agent against fungi. This study aimed to isolate and characterize chitinase-producing bacteria from contaminated soil and to determine their potential as biocontrol agents against Penicillium sp. Chitinolytic bacteria were isolated using Nutrient Agar (NA) medium containing colloidal chitin, followed by morphological and biochemical characterization. Enzyme production was carried out using medium containing colloidal chitin, while chitinase activity was determined using the DNS method by measuring absorbance at a wavelength of 540 nm. The results showed that two chitinolytic bacterial isolates, namely E105a and E105b, were obtained. Based on the enzyme activity assay at pH 7 and a temperature of 37°C, isolates E105a and E105b showed enzyme activities of 0.0371 U/mL and 0.0302 U/mL, respectively. The potential of chitinase as a biocontrol agent against Penicillium sp. was demonstrated by inhibition percentages of 47,30% for isolate E105a and 43,56% for isolate E105b.
