Abstract
Industri pengecoran logam merupakan salah satu sumber limbah yang mengandung logam berat timbal (Pb). Desa Jeblokan, Kecamatan Ceper merupakaan salah satu lokasi sentra pengecoran logam di Kabupaten Klaten. Akumulasi kandungan timbal di lingkungan sekitar pengecoran logam akan menimbulkan beberapa gangguan kesehatan sehingga diperlukan upaya-upaya untuk mengatasinya. Salah satu alternatif yang dapat dilakukan adalah dengan memanfaatkan kemampuan mikroba dalam melakukan remidiasi logam berat timbal. Langkah awal yang dapat dilakukan adalah dengan mengetahui tingkat toleransi mikroba untuk tumbuh pada lingkungan yang mengandung logam berat Pb. Tujuan penelitian ini adalah untuk mengetahui kemampuan toleransi bakteri yang diperoleh dari tanah bekas cetakan pengecoran logam terhadap logam berat Pb. Media yang digunakan untuk isolasi bakteri adalah media zobell 2216 E agar yang mengandung timbal 0,1 ppm. Screening untuk menentukan tingkat kemampuan bakteri yang toleran terhadap Pb menggunakan media zobell dengan variasi konsentrasi timbal 0,1 ppm; 0,2 ppm dan 0,3 ppm. Isolat unggul yang mampu tumbuh pada konsentrasi Pb tertinggi dikarakterisasi secara fenotipik dan diidentifikasi menggunakan metode profil matching. Hasil isolasi bakteri diperoleh 10 isolat. Terdapat 3 isolat unggul yang memiliki kemampuan tumbuh pada konsentrasi Pb tinggi (0,3 ppm) yaitu isolat P5, P6 dan P7. Berdasarkan hasil identifikasi isolat P5, P6 dan P7 masing-masing termasuk anggota genus Enterococcus, Lactobacillus dan Pseudomonas.
