Abstract

River is a water source that are useful for human needs. Human activities in using river water and dumping waste to the river could decrease water quality of the river. This research was intended to find out the MPN index of coliform bacteria and coliform bacteria spesies diversity as bioindicator of water quality in Gajah Wong river, DIY, and the physical parameter (temperature, turbidity, velocity flow) and chemical parameter (pH, DO, BOD). Water sampling was conducted at three location: upstream at Hargobinangun, midstream at Rejowinangun, and downstream at Pandeyan. The sample was taken from six point from each location and the distance between the point was 20 meter. The coliform bacteriological testing used MPN (Most Probability Number) method including presumptive test, confirmed test, and a completed test, and continued with identification using IMViC test, hydrogen sulfide production, motility, and urea test. Based on the standard of water quality Ordinance Government Republic of Indonesia Number 82 Year 2001 about Water Quality Management and Water Pollution Control, and also Ordinance of the Minister of Health Republic of Indonesia Number 416/MENKES/PER/IX/1990, water quality of Gajah Wong river included as in class III and IV that classified into moderately to heavily polluted. The microbiology analysis result showed that Gajah Wong river contains five coliform bacteria group, they were Escherichia coli, Providencia sp., Morganella morganii, Proteus vulgaris, and Proteus mirabilis. Based on Correlate Bivariate analysis, the MPN have a correlation with the velocity flow, and the bacteria diversity have a correlation with BOD.
